The Expression and Tissue Localization of Kir6.2 gene in the pancreas of animal models for non-insulin-dependent diabetes

Abstract

|
ATP-sensitive potassium(Kir6.2) channels play an important role in the secretion of insulin. So we have examined the expression and distribution of Kir6.2 gene in pancreas of NIDDM mouse model, C57BL/KsJ-Lepr^(db/db). The distribution of Kir6.2 mRNA expression were determined by in situ hybridization with digoxigenin(DIG)-labeled antisense cRNA probe. The hybridization signal was evenly present throughout pancreatic islets in control C57BL/6 mouse(B6), while in C57BL/Ks-Lepr^(db/db) mouse, hybridization signal was mainly detected in alpha-cell region of the pancreatic islets. And the differentiation of Kir6.2 mRNA expression between C57BL/KsJ-Lepr^(db/db) and B6 mouse was investigated caning out the Ribonuclease Proteciton Assay(RPA) using by P^(32)-labeled antisense riboprobe. We found that the expression of Kir6.2 gene decrease(22.3%) in the pancreas of C57BL/KsJ-epr^(db/db) mouse compared with B6 mouse. The reduced Kir6.2 expression is related to the insulin secretory response of ¥â-cell to elevated blood glucose. These results suggest that Kir6.2 is an important component in the regulation of the release of both insulin and glucagon.
